The sinkhole was reported around 11 p.m. Wednesday at Morena Boulevard and Sherman Street, according to the San Diego Police Department.
Police said the closure on southbound Morena was expected to continue throughout the day, with one lane on northbound Morena that remained open.
The cause of the sinkhole was unclear, but a water main break was also reported in the Morena area on Thursday morning.
No injuries were reported.
