Born in June 1925 in New York City, Lockhart was the daughter of Broadway actor Gene Lockhart and English-born actress Kathleen Lockhart. She started her career on stage at just eight years old, playing Mimsey in Peter Ibbetson, presented by the Metropolitan Opera. Her film and TV career didn't start until 1938, though, when she appeared with her parents in the film A Christmas Carol. Six years later, she starred in the 1944 musical Meet Me in St Louis alongside Judy Garland, Margaret O'Brien, Mary Astor, Lucille Bremer, Tom Drake, Leon Ames, Marjorie Main and Joan Carroll.

In 1948, Lockhart won a Tony Award for her role on Broadway in For Love or Money. She later donated her award to the Smithsonian Institution in 2008 for display in the museum's entertainment archives.

From 1958 to 1964, Lockhart portrayed the mother of Timmy Martin (Jon Provost) in the CBS series Lassie, where she was nominated for an Emmy Award twice. Then, Lockhart played Dr. Maureen Robinson in Lost in Space, which ran from 1965 to 1968 on CBS, opposite Guy Williams and Jonathan Harris. Afterward, she appeared as Dr. Janet Craig on the final two seasons of the CBS sitcom Petticoat Junction from 1968 to 1970.

Throughout the 1970s up until she retired from acting in 2021, Lockhart mostly just made guest appearances and had small roles on various shows and movies, including Butterfly, General Hospital, Bewitched, Family Affair,Roseanne, Full House, Grey's Anatomy and many more. Her last active role was lending her voice in the 2021 Lost in Space reboot as June, or the Voice of Alpha Control.
