Laurel County Sheriff John Root says deputies were originally called for a report of a gunshot victim around 5:30 p.m. Saturday on I-75 near exit 49.
When deputies arrived, they found nine vehicles had been shot into in both the north and southbound lanes. Five people were found to be seriously injured.
Officials have identified Joesph A. Couch, 32, of Woodbine, Ky as a person of interest in the case.
Couch is considered armed and dangerous and should not be approached.
The scene is being contained and the investigation will continue at daybreak on Sunday morning.
Multiple agencies are assisting on the investigation.
Andy Beshear said he, alongside the Kentucky State Police and the Office of Homeland Security, "are actively monitoring the situation and offering support in any way possible."
Laurel County Sheriff's Office is leading the investigation with assistance from several other law enforcement agencies, including Kentucky State Police, Pennington said.
The Bureau of Alcohol, Tobacco, Firearms, and Explosives also confirmed it was involved in the search.
Earlier, Kentucky's Governor Andy Beshear wrote on social media that law enforcement had "shut the interstate down in both directions" near to where the shooting happened.
He said the area should be avoided, and urged the public to "pray for everyone involved".
London is a small city of about 8,000 residents near the Daniel Boone National Forest.
